Molecular Biology & Biochemistry - Biol 99        Homework 3    Due March 11, 2008

Using the textbook, Wikipedia, and any other sources you may choose (although it is not necessary to go further, you must at least use these two sources),

1. List the enzymes, reagents, and special conditions required for PCR, the polymerase chain reaction, and briefly (a sentence or less) explain their specific functions or roles in PCR.

2. Briefly describe and diagram PCR.  Be sure that every item listed in part 1 is included in this diagram.  The recommended way to answer this is a flow diagram, with annotations that explain the steps as necessary.  In some cases this may simply be a reference to part 1 components (no need to repeat yourself).

3.  Describe and explain three distinct uses of PCR.

4.  What sorts of DNA might be used as primers?  For each of the uses you have described in #3, explain what sort of primer, or the characteristics of the primers that would be used.

The flow diagram  may be written by hand, but at least parts 1 and 3 must be printed (computer).  If you write anything by hand be sure that it is easily legible.  If you have any doubts, use the computer!

You may use any source(s) you wish, but do not simply copy/paste diagrams - you may copy BY HAND or use the computer to generate one YOURSELF.  You should be able to recreate this diagram or flowchart without notes, so be sure to practice.
